Abstract:
The present invention relates to the field of detecting and quantifying apoptosis. In one aspect, the invention is directed to an isolated monoclonal or polyclonal antibody that specifically recognizes caspase-generated products of K18 or K19 but does not react with intact K18 or K19. The antibody typically reacts with the cleavage site of K18 or K19 at Asp237. The binds to the epitope common to caspase-cleaved K18 and K19-(T/S)VEVD- and the Asp-proximal valine is essential for antibody recognition.

Abstract:
The present invention is generally directed to biological assays. More specifically it is directed to FRET-based assays using particularly effecting FRET pairs, methods for performing such assays and the molecules utilized in the assays. In a composition aspect, the present invention is directed to a FRET-based protease substrate selected from the list of substrates shown in the Detailed Description section above. In a method aspect, the present invention is directed to a method of performing a FRET-based assay, where the assay includes the following steps: adding a solution or suspension containing one or more different proteins to a reaction vessel; adding a liquid comprising a FRET-substrate to the reaction vessel, wherein the FRET-substrate is selected from the list of substrates shown in the Detailed Description section above. Abstract:
A method for isolating D and L amino acids from a racemic mixture of the formula (DL)--H. sub. 2 N--CHX--COOY, and salts thereof, wherein X denotes an amino acid side chain and derivatives thereof and Y denotes a suitable alkyl group. The racemic mixture is contacted with water to form an aqueous solution. This aqueous solution is then contacted with an esterase to thereby obtain a solution comprising the resolved amino acids (L)--H. sub. 2 N--CHX--COOH and (D)--H. sub. 2 N--CHX--COOY. The N-terminus of the resolved (L) and (D) amino acids present in this solution are protected with an N-protecting group R to thereby obtain a solution comprising (L)--R--NH--CHX--COOH and (D)--R--NH--CHX--COOY. The isolation of (L)--R--NH--CHX--COOH from (D)--R--NH--CHX--COOY is achieved in high yields by an acid-base extraction.
